Abstract

Objective: Baclofen is a non-selective antispasmodic agent used to treat spinal disorders in patients with multiple sclerosis. Its use in spasticity has been limited by the lack of efficacy and side effects. We sought to assess the safety and efficacy of baclofen in treating spasticity in patients with multiple sclerosis. Methods: This single-arm, double-blind, randomized, placebo-controlled study included 50 patients with multiple sclerosis, with or without spasticity. All patients were treated with a spasticity control medication, baclofen. The primary efficacy endpoint was the change in spasticity severity. The secondary endpoints were improvement of spasticity scores with the Baclofen Symptom Schedule (BSSS) and the improvement in functional independence with the Baclofen Questionnaire (BLQ). Results: The BSSS demonstrated a significant reduction in spasticity scores in patients who experienced improvement with the Baclofen Questionnaire compared to those who experienced the improvement in spasticity scores in the placebo group. The BLQ demonstrated a significant reduction in the severity of spasticity score improvement with the Baclofen Symptom Schedule (BSSS). The BSSS demonstrated improvement in functional independence with the Baclofen Questionnaire compared to improvement in the placebo group.

Primary efficacy outcomes

The BSSS demonstrated improvement in spasticity scores for all patients at the treatment endpoint. These improvements were more pronounced in patients who experienced improvement with the Baclofen Questionnaire compared to patients who experienced improvement in the placebo group. There were no significant differences in the scores between the BSSS and BLQ patients.
